if I have 2 gb ram in one slot,then I can only use 2 gb ram on second slot or 4gb will also work..
and can someone suggest ram for lenovo g500 laptop..

If you are using 32-bit OS, then, your CPU will address up to 4GB only (Including GPU Memory), regardless to the amount of RAM you have in your system. Switch to 64 bit OS
You can mix any quantities of RAM, it will add up, but one thing if your mother board supports, if you have two identical RAM sticks in both slots, your system will use dual channel configuration to increase 2XBand width of RAM, if there are any unidentical RAM sticks in slots, then dual channel will not be enabled but quantity wise you will not get any issue
